You can buy magnetic vent covers for your vents (provided they are made of metal) that will completely block them off. I linked to Amazon so you can see what I'm talking about but they should have them at any Walmart or Home Improvement/Hardware store.

I’ve only seen them for the rectangular ones.
My vent is more of a square (9” x 8.5”) so those won’t work. But thank you for the suggestion! I’ll probably end up covering the vent with a mixture of foam and moving blanket to muffle the sound of my roommate.

The ducts that go from the AC unit to the registers will have a considerable amount of surface area. If it goes through the attic, it will gather a relatively substantial amount of heat. Any small difference in pressure will cause that heat to enter the house through the vent.
If he is using a window unit or something similar, he is correct in wanting to cover the register. This will save him money in relatively short order. (assuming he is in a hot area)
I would recommend anyone covering the vents of a system that is not in use. This will help insulate from the exchange that will occur in your duct system.
