The backplate is very easy to remove. You need a triwing screwdriver to do it, but it's pretty straightforward. There's also a protective metal plate just under the plastic, so there's not much room to mess up anything. I replaced the shell on mine and had no issues (joycons were a bit rough, though). Here's a video on removing it.

I second the flex cable idea. You’ll need a triwing and jeweler’s screwdriver, some small sorting containers (ice cube tray works great), and some patience. The screen connector is held in with a little flap tab, make sure contacts are clean for both male and female sides (isopropyl alcohol rub down, allow to completely dry in open air after cleaning). Also ensure connector is fully and evenly inserted. Inspect cable for kinks, breaks, etc.
The only other relatively cheap and easy fix is screen capactor or resistor reflow/replacement, if you already have soldering equipment and experience. Otherwise, not much else you can do but replace the screen.

2,4,5. Your best bet is soap and warm water or some other house hold cleaner. I use a toothbrush, you could use a cotton swab instead. I read that isopropyl alcohol damages/discolors the controller if overused. I use it anyway with cotton swabs for all the nasty bits. If you want to clean the the shoulder buttons, you will also need a Phillips screwdriver to remove the shield.
In terms of difficulty, your only problem is losing the buttons, pads, and screws. I dip my parts in soap water and scrub with a toothbrush, that’s it.
If you want to clean the stick box itself. I just use rubbing alcohol and a cotton swab to clean up whatever is exposed and dirty. As for the inside, you will likely need a soldering iron. Refer to the guide for disassembly and maintenance.
