EDIT: removed IDW's Transformers
Start here. It's not the first volume (that's published by Fantagraphics and is collected in a $75 omnibus that's definitely worth checking out if you find you do like the series) but it's a good jumping on point and over 600 pages of content for an MSRP of $24.99 is pretty hard to beat.
The first chapter of the first volume of Usagi Yojimbo features the Turtles in feudal Japan. It's a really fun read.

Usagi is essentially 33 trades and then the new Issue #1 from IDW. These are almost all available in cheap omnibus collections, though.
Fantagraphics has a 2 part omnibus collection Usagi Yojimbo: The Special Edition. This covers the first 7 volumes of the book.
